"Generation of xenochimeric blastoids assembled from human expandable trophectoderm-like cells and mESCs"

ABSTRACT
Trophoblast cells are essential for embryonic development. However, current human trophoblast stem cells (hTSCs) do not faithfully recapitulate trophoblast development over the pre- and peri-implantation stages. To address this, we established human trophectoderm-like stem cells (hTELSCs) from pluripotent preimplantation epiblast stem cells. Human TELSCs can be expanded for more than 20 passages and share molecular features with trophectoderm but differ from post-implantation cytotrophoblasts and hTSCs. Human TELSCs can be developed into cytotrophoblast, syncytiotrophoblast and extravillous trophoblast lineages. Importantly, hTELSCs efficiently self-organize into xenochimeric blastoids (xeno-blastoids) when partnered with mouse pluripotent stem cells. The human-mouse xeno-blastoids are capable of implantation and inducing decidualization of mouse endometrium. Overall, hTELSCs provide a powerful tool to study human blastocyst development, implantation, and placentation.

Jichang Wang (王继厂), currently Professor of Histology and Embryology in Sun Yat-sen University (SYSU), Guangzhou, P.R. China. His main research interest is to elucidate the function of retrotransposons (e.g., endogenous retrovirus, LINE-1) in mammalian early development and neurodevelopmental diseases by using stem cells and interspecies chimeras as models and by integrating genome editing and multi-omics technologies. Dr. Wang is supported by the National Science Fund for Distinguished Young Scholars, the National Key Research and Development Program of China, and the National Science Foundation of China. He was a winner of the “Publication of the Year” award of German Stem Cell Network (2015). He has published many papers in Nature, Cell Stem Cell, PNAS, Trends in Microbiology, Nature Protocols, Cell Reports, Nucleic Acids Research, Genome Biology, etc.

"Naive human PSCs model pre- to post-implantation development"

ABSTRACT
We aim to understand early human development by focusing on the pre-implantation to post-implantation stages. To achieve our objective, our group has established a series of methods and human cells that correspond to human peri-implantation embryos. The blastocyst contains three cell types, epiblast, hypoblast, and trophectoderm. We successfully induced hypoblast and trophectoderm of the pre-implantation stage from naive human pluripotent stem cells (PSCs), which share features with pre-implantation embryos. In this presentation, I will present in vitro lineage specifications for human extraembryonic lineages from naive human PSCs and in vitro development using naive PSCs together with hypoblast-like cells and trophectoderm-like cells during peri-implantation.



BIO
Yasuhiro Takashima is a professor at Kyoto University, CiRA. Takashima, a graduate of Kobe University School of Medicine, transitioned from internal medicine practice to research. After he earned his Ph.D at Kobe University Graduate School and RIKEN Kobe, focusing on adipocytes and mesenchymal stem cells using mouse embryonic stem cells, he worked at the University of Cambridge Stem Cell Institute as a postdoctoral fellow, making significant contributions like the establishment of naive human pluripotent stem cells. Since 2015, Takashima and his Lab aims to regulate stem cells for the promotion of human health, with a particular focus on early human development and regenerative medicine using pluripotent stem cells and tissue stem cells at Kyoto University CiRA.
